我用TAE跑電泳..
也是用EtBr染色...
在用紫外線照就可以看見band..
不過要注意..
配agarose gel用的溶劑要和buffer一樣..
向用0.5X TAe跑配的時候就要用0.5XTAE..
不然會跑不出來...

跑了快十年的 TAE agarose 電泳
沒聽說過這樣的說法
你的 product 大小有多少

TAE 在大部份的 agarose 電泳均可
但缺點是跑太久會 pH 會跑掉就會出狀況
也有人死不肯換但沒啥太大問題
TBE 比 TAE 來的貴 (成本上)
但在看小分子量的時候可以讓 band 的解像力比較好
也有人資本雄厚所有的電泳一種 TBE 玩到底
並且跑長時間 pH 不易變化
很多長時間的電永都用這種 buffer
你的產物若是不大
選 PAGE 用 TBE 來跑應該不錯 (20bp-90bp) 應該不錯
若是大於 200-500 bp 用 TAE 4% agarose EtBr 預先加在 gel 中
也可以清楚看到
但大於 1kb 以上 1% agarose TAE 跑完放在 EtBr 的染缸染一陣子就一定沒問題了

通常用是用一樣的buffer....
可是如果用不一樣的還是可以做得出來
有一國的說法是：
如果你用1X TAE gel 配合 0.5X TAE buffer...
因為buffer的離子量相對較少
你的DNA sample反而可以跑得比較快
因為外面的電阻較大
聽起來很有道理吧？
不過我想是聽聽就好。
我只是要說：
沒有什麼會是做不出來的
只要會導電，也許要等，可是還是做得出來。

就是結果很怪就對了...

這可不一定,我曾這樣跑過,但沒問題~(我是自己去 test 的)

我的意思不是說你可以用water gel來做
因為water跟buffer 比起來電阻又要大得多
而且你的sample在Gel中反而會變成
被導電性不良的gel 繞著
我想這會是很大的問題....
如果你用的伏特很高的話也許會把你的Gel熔掉
而且...
哦～如果你有看清礎的話
我說的是用1X TAE gel + 0.5 buffer...
在Gel中的buffer 比外頭要高。
看有沒有人有空來做這個實驗....
我本來的意思是，別的方法還是可以用
不過當然平常人都是用一比一，
除了方便以外
防止不同濃度的buffer exchange影響結果
當然也可能是原因

如果不行，gel用的buffer濃度不能比電泳用buffer濃度低....

這可不一定,要看你買的是那一家的電泳槽.
像 COSMO BIO CO., 的 Mupid-2 就建議用 0.5X 的 buffer 來跑,
gel 還是用 1X 的....所以多看看 說明書吧~
